Research in our laboratory is focused on the early pathogenetic events driving diffuse large B-cell lymphoma (DLBCL) and in the immune evasion strategies of this most common and lethal type of B-cell lymphoma. Data generated in our lab implicate regulatory T-cells (Tregs) as an attractive target for immunotherapy in a subset of DLBCL patients. However, the precise role of Tregs in lymphomagenesis is largely unknown. We have discovered that some genetically defined subtypes of DLBCL are infiltrated by large numbers of Tregs. This finding is recapitulated in experimental animal models of the disease. By harnessing single cell RNA-sequencing and high-dimensional flow cytometry approaches we find that lymphoma-infiltrating Tregs differ strongly from naive Tregs residing in the corresponding lymphoid organs of healthy mice, both in terms of their transcriptional signature and immunophenotype. The depletion of Tregs in a transgenic model abrogates lymphoma growth, and the addition of Tregs to lymphoma cell cultures boosts their proliferation ex vivo. Our data suggest that Tregs support lymphoma B-cells beyond suppressing anti-lymphoma immunity.

**Your responsibilities**:
You will be working with experimental mouse models of DLBCL that include syngeneic orthotopic tumor models and genetically humanized immunodeficient mouse models. You will explore the tumor B-cell/Treg interface in vitro and in vivo using co-culture systems, microscopy, and high-dimensional flow cytometry. You will test various Treg-directed interventions as promising novel treatment strategies, both alone and in combination with established chemo
- or targeted therapies. You will further use existing patient cohorts and patient datasets to investigate how Treg infiltration in human DLBCL correlates with mutational signatures, the clinical history and prognosis of the patient on standard therapy, and other hallmarks of the disease. Supervising block course and/or master students will be part of your responsibilities within the graduate school.

**Your profile**:
We are seeking to hire a PhD student with a practical and theoretical background in immunology and/or cancer biology to complement our team. Basis experience in flow cytometry, cell culture, and animal experimentation are a plus but not a prerequisite.
