**PhD position**
**60 % **:The Institute of Pharmacology and Toxicology is an experimental institute of the Faculty of Medicine and the Faculty of Science of the University of Zurich. The scientific focus is on practical research into diseases of the nervous system. The Department of Neuropharmacology, under the direction of Prof. H.U. Zeilhofer, uses state-of-the-art methods of neuroscience (optogenetics, chemogenetics, viral gene transfer, etc.) to study the spinal mechanisms of pathological pain mainly in mouse models. The majority of the research projects are based on the use of genetically modified mice.

**Your responsibilities**: Pain, especially chronic pain, is a major health issue affecting millions, with many still

lacking appropriate treatment. New therapeutic avenues will likely emerge with a better understanding of the neuronal mechanisms underlying the sensory, emotional and cognitive aspects of pain as well as their interaction. While the spinal cord conveys sensory information to the brain, it also receives input from several brain regions involved in nociception. This descending input can affect spinal sensory processing, resulting in a spinal-brain-spinal sensory loop. This PhD project aims to disentangle the contributions of the spinal cord and the brain to nociceptive signaling in mice. For example, how does the state of an animal (anesthesia, sensory expectation, chronic pain...) influence spinal output to the brain? To address this question, the PhD student will take advantage of cutting-edge neuroscience tools to measure spinal cord neurons activity in vivo with single cell resolution.

This PhD position provides a unique opportunity to acquire expertise in state-of-the-art neuroscience techniques including:

- stereotactic surgeries including adeno-associated viruses injection to the brain and the spinal cord, implantation of chronic imaging windows, and implantation of optic fibers for optogenetics
- in vivo two-photon imaging
- neuronal activity recording from behaving mice carrying with a miniature microscope
- advanced behavioral experiments and neuronal data collection
- circuit neuroscience techniques to modulate the activity of specific neuronal populations (optogenetics, chemogenetics)
- data analysis and programming
- hold a MSc or equivalent in neuroscience or related field
- excellent English speaking and writing skills
- good understanding of basic neuroscience
- basic programming skills (Python, Matlab, R...) preferred
